Alkanes and alkenes are high-value platform chemicals that can be synthesized by microorganisms, utilizing organic residues from agri-food industries and municipalities, thereby offering an alternative opportunity in resource recovery. Current research and technological advancements for the biosynthesis of alkanes and alkenes are mainly impeded by low product titers, obstructing the bioprocess upscaling and large-scale applications. Thus, current scientific investigations aim to improve productivity by utilizing natural and engineered metabolic pathways in various microbial chassis to suppress competing metabolic pathways, coupled with bioprocess optimization. Additionally, to reduce costs, research is being conducted on utilizing inorganic carbon sources such as CO2 to promote the green synthesis of alkanes and alkenes. Therefore, this review critically discusses the opportunities and challenges in alkane and alkene biosynthesis, aiming to examine the current technological advancements. In this review, the limitations of five major metabolic pathways for alkane and alkene biosynthesis are thoroughly discussed, highlighting their shortcomings. Additionally, various techniques, including metabolic engineering, autotrophic metabolic pathways, and new non-biosynthetic routes, are investigated as potential methods to enhance product titers. Furthermore, this review offers valuable insights into the economic and environmental aspects of alkane and alkene biosynthesis while also presenting perspectives for future research directions.

Abstract Galaxaura divaricata is a partially calcified macroalga that hampers coral recruitment, growth, and recovery via the excretion of allelopathic secondary metabolites. Herbivorous fishes are not major consumers of Galaxaura spp. and there is a need to understand feeding preferences for Galaxaura divaricata in other macroherbivores, like sea urchins and green sea turtles that could act as potential controlling agents. Under certain environmental conditions, G. divaricata can proliferate and overgrow degraded reefs for several years, as documented for several coral patch reefs in the lagoon of Dongsha Atoll, South China Sea. This study aimed to experimentally test the feeding preferences of five species of sea urchin and two individual green sea turtles, Chelonia mydas, for G. divaricata. Specifically, we quantified and compared the consumption rates of the allelopathic G. divaricata with Gracilaria edulis, a nonallelopathic, fleshy red alga, known to be highly favored by herbivores. Results showed that the five urchin species fed on both G. edulis and G. divaricata. However, urchins consumed 2–8 times less wet weight of G. divaricata (range 0.3–3.1 g urchin−1 24 h−1) compared to G. edulis (range 0.6–18 g urchin−1 24 h−1), suggesting that urchin grazing may exert some control on G. divaricata abundance but is likely ineffective for a large‐scale removal of the alga. Further, both green sea turtles avoided G. divaricata and selectively fed on G. edulis. More experiments are needed to test the potential role of herbivores in controlling the overgrowth of coral competitive and allelopathic macroalgae, like Galaxaura on coral reefs.

Abstract Purpose The aged microenvironment plays a crucial role in tumor onset and progression. However, it remains unclear whether and how the aging of the extracellular matrix (ECM) influences cancer onset and progression. Furthermore, the mechanisms and implications of extracellular matrix senescence-related genes (ECM-SRGs) in pan-cancer have not been investigated. Methods We collected profiling data from over 10,000 individuals, covering 33 cancer types, 750 small molecule drugs, and 24 immune cell types, for a thorough and systematic analysis of ECM-SRGs in cancer. Results We observed a significant correlation between immune cell infiltrates and Gene Set Variation Analysis enrichment scores of ECM-SRGs in 33 cancer types. Moreover, our results revealed significant differences in immune cell infiltration among patients with copy number variations (CNV) and single nucleotide variations (SNV) in ECM-SRGs across various malignancies. Aberrant hypomethylation led to increased ECM-SRGs expression, and in specific malignancies, a connection between ECM-SRGs hypomethylation and adverse patient survival was established. The frequency of CNV and SNV in ECM-SRGs was elevated. We observed a positive correlation between CNV, SNV, and ECM-SRGs expression. Furthermore, a correlation was found between the high frequency of CNV and SNV in ECM-SRGs and poor patient survival in several cancer types. Additionally, the results demonstrated that ECM-SRGs expression could serve as a predictor of patient survival in diverse cancers. Pathway analysis unveiled the role of ECM-SRGs in activating EMT, apoptosis, and the RAS/MAPK signaling pathway while suppressing the cell cycle, hormone AR, and the response to DNA damage signaling pathway. Finally, we conducted searches in the “Genomics of Drug Sensitivity in Cancer” and “Genomics of Therapeutics Response Portal” databases, identifying several drugs that target ECM-SRGs. Conclusions We conducted a comprehensive evaluation of the genomes and immunogenomics of ECM-SRGs, along with their clinical features in 33 solid tumors. This may provide insights into the relationship between ECM-SRGs and tumorigenesis. Consequently, targeting these ECM-SRGs holds promise as a clinical approach for cancer treatment.

Background: This study focussed on exploring the prognostic prediction performance of the growth arrest and DNA damage-inducible 45 beta (GADD45B) and its associations with T-cell activity and immune soakage in different malignancies, especially lung squamous cell carcinoma (LUSC). Methods: We applied TIMER database for comparing the expressions of GADD45B among different cancers. OncoLnc, Gene Expression Profiling Interactive Analysis 2 (GEPIA2), and Kaplan-Meier Plotter were utilised to evaluate the prognostic prediction performance of GADD45B. Besides, the associations of GADD45B with clinical stage, associated gene markers, and immune infiltration were examined through TISIDB, GEPIA2, and Tumour Immune Estimation Resource (TIMER). Biological processes (BPs) and KEGG enrichment analyses were performed to illustrate the possible role of GADD45B in LUSC. The miRWalk database was adopted to analyse the gene miRNA interaction network of GADD45B in LUSC. Results: GADD45B expression was decreased in most of the malignancies, with relation to the poor prognosis in LUSC. GADD45B also significantly affected the survival of LUSC subgroups divided by clinic data. GADD45B significantly correlates with and may stimulate T cell exhaustion in LUSC. Conclusions: GADD45B is a prognostic indicator in multiple tumours, especially in LUSC. Moreover, modulating GADD45B expression may improve immunotherapy efficacy in LUSC.

Purpose: To predict the shelf life of milk powder based on the change of oxygen concentration in the packaging headspace. Methods: The relative oxygen consumption rate was used to characterize the oxidation characteristics of whole milk powder, and the oxygen consumption threshold was determined by analyzing several oxidation indexes. Oxygen permeability of the packaging materials and oxidation characteristics of milk powder were integrated to characterize the change of headspace oxygen concentration, and a shelf life prediction model of milk powder was established based on the total oxygen consumption. The model was verified by accelerated storage test of polyethylene terephthalate/polyethylene packaged whole milk powder at 45 ℃. Results: The oxygen consumption threshold of whole milk powder was 0.041 47 mg/g. The experimental data of oxygen concentration in the packaging headspace during accelerated storage was consistent with the predicted data, and the relative deviation between the experimental and theoretical shelf life was 6.06%. Conclusions: The shelf life of whole milk powder can be predicted by the change of oxygen concentration in the packaging headspace.

Abstract Oilseed rape (Brassica napus) is an important crop that is cultivated for the oil (mainly triacylglycerol; TAG) it produces in its seeds. TAG synthesis is controlled mainly by key enzymes in the Kennedy pathway, such as glycerol 3-phosphate acyltransferase (GPAT), lysophosphatidate acyltransferase (LPAT) and diacylglycerol acyltransferase (DGAT) but can also be produced from phosphoglycerides such as phosphatidylcholine (PC) by the activity of the enzyme phospholipid: diacylglycerol acyltransferase (PDAT). To evaluate the potential for these enzymes to alter oil yields or composition, we analysed transgenic B. napus lines which overexpressed GPAT, LPAT or PDAT using heterologous transgenes from Arabidopsis and Nasturtium and examined lipid profiles and changes in gene expression in these lines compared to WT. Distinct changes in PC and TAG abundance and spatial distribution in embryonic tissues were observed in some of the transgenic lines, together with altered expression of genes involved generally in acyl-lipid metabolism. Overall our results show that up-regulation of these key enzymes differentially affects lipid composition and distribution as well as lipid-associated gene expression, providing important information which could be used to improve crop properties by metabolic engineering.

Disulfidptosis, a novel form of regulated cell death (RCD) associated with metabolism, represents a promising intervention target in cancer therapy. While abnormal lncRNA expression is associated with colon cancer development, the prognostic potential and biological characteristics of disulfidptosis-related lncRNAs (DRLs) remain unclear. Consequently, the research aimed to discover a novel indication of DRLs with significant prognostic implications, and to investigate their possible molecular role in the advancement of colon cancer. Here, we acquired RNA-seq data, pertinent clinical data, and genomic mutations of colon adenocarcinoma (COAD) from the TCGA database, and then DRLs were determined through Pearson correlation analysis. A total of 434 COAD patients were divided in to three subgroups through clustering analysis based on DRLs. By utilizing univariate Cox regression, the least absolute shrinkage and selection operator (LASSO) algorithm, and multivariate Cox regression analysis, we ultimately created a prognostic model consisting of four DRLs (AC007728.3, AP003555.1, ATP2B1.AS1, and NSMCE1.DT), and an external database was used to validate the prognostic features of the risk model. According to the Kaplan–Meier curve analysis, patients in the low-risk group exhibited a considerably superior survival time in comparison to those in the high-risk group. Enrichment analysis revealed a significant association between metabolic processes and the genes that were differentially expressed in the high- and low-risk groups. Additionally, significant differences in the tumor immune microenvironment landscape were observed, specifically pertaining to immune cells, function, and checkpoints. High-risk patients exhibited a low likelihood of immune evasion, as indicated by the Tumor Immune Dysfunction and Exclusion (TIDE) analysis. Patients who exhibit both a high risk and high Tumor Mutational Burden (TMB) experience the least amount of time for survival, whereas those belonging to the low-risk and low-TMB category demonstrate the most favorable prognosis. In addition, the risk groups determined by the 4-DRLs signature displayed distinct drug sensitivities. Finally, we confirmed the levels of expression for four DRLs through rt-qPCR in both tissue samples from colon cancer patients and cell lines. Taken together, the first 4-DRLs-based signature we proposed may serve for a hopeful instrument for forecasting the prognosis, immune landscape, and therapeutic responses in colon cancer patients, thereby facilitating optimal clinical decision-making.

Abstract In degraded coral reef ecosystems, allelopathic macroalgae have received increasing attention from marine ecologists because their secondary metabolites (also known as allelochemicals) kill corals that grow adjacent to them and weaken the recovery of degraded reefs. One well‐known coral‐killing macroalga is the calcareous red seaweed Galaxaura. However, our knowledge of how coral reef fishes interact with allelopathic algae like Galaxaura is very limited. Here, we documented novel observations of feeding interactions of 17 species of coral reef fishes (herbivorous and carnivorous) with the filamentous Galaxaura divaricata on degraded lagoon patch reefs in Dongsha Atoll (South China Sea). Video analyses showed that territorial farming damselfishes (i.e., Dischistodus perspicillatus, D. prosopotaenia, Hemiglyphidodon plagiometopon, Pomacentrus grammorhynchus, P. adelus, and Neoglyphidodon nigroris) and juvenile parrotfishes (Scarus schlegeli, S. ghobban, S. rivulatus, and Chlorurus spilurus) likely used G. divaricata as a feeding substratum. Further, microscopic analyses revealed that the filamentous surface of G. divaricata harbored a wealth of epiphytic microalgae, such as filamentous cyanobacteria (i.e., Leptolyngbya, Lyngbya, Rivularia, Oscillatoria, and Stigonema), diatoms (i.e., Synedra, Nitzschia, Mastogloia, and Pleurosigma), and filamentous red algae (i.e., Heterosiphonia), suggesting that these fishes targeted the nutrient‐rich microscopic epiphytes rather than the nutrient‐poor host. Juvenile benthic carnivores (i.e., Labridae, Parupeneus multifasciatus, and Meiacanthus grammistes) form feeding assemblages with roving parrotfishes to feed on small invertebrates (i.e., amphipods, copepods, isopods, gastropods, and polychaetes) associated with G. divaricata. Given that coral reef fishes appear to target the epiphytes associated with Galaxaura rather than the alga itself, these observations thus substantiate the threat posed by the overgrowth of G. divaricata to coral recovery in degraded reef systems due to the lack of natural grazers.

Untethered magnet‐driven microrobots play an increasingly important role in various biomedical applications. Incorporating bionic technology into microrobot design is an emerging way to improve the work efficiency of microrobots. Herein, a magnetically powered and frictional anisotropy‐based microwalker that can be potentially used in in vivo nonliquid‐filled environment is proposed. The microwalker is constructed by two rigid segments with an equal length of 70 μm, connected by a rigid joint. Parallel gecko setae‐like tentacles are placed at the bottom of the segments as contact feet to generate friction with the contact surface. The microwalker is integrally fabricated from biocompatible materials with 3D laser lithography based on two‐photon polymerization. The microwalker can be well controlled to move in low‐Reynolds (Re)‐number regimes under an external oscillating magnetic field. In addition to moving in a liquid environment as existing microswimmers, the microwalker can move in surface in a nonliquid‐filled environment. It can also climb the slope driven by the planar magnetic field only. Several experiments were conducted to demonstrate good motion capability of the microwalker. This study provides a new solution to microrobot design for future biomedical applications.

Regenerated cellulose membrane is a biomaterial obtained by activating, dissolving, solidifying, and regenerating cellulose powder using solvents of different polarities. It has the characteristics of high oxygen resistance and high strength. However, its low water vapor barrier and single function limit its application. In order to improve the water resistance of regenerated cellulose membranes and endow them with UV resistance, lignin was extracted from waste wheat straw using formic acid method. The extracted formic acid lignin (FAL) was added to the cellulose solution to prepare a series of regenerated lignocellulosic membranes (RC‐FAL) with different lignin contents. The results indicate that lignin can not only improve the water vapor barrier, tensile strength, and water resistance of the film, but also enhance the oxygen barrier and UV absorption of the film. Compared with pure cellulose film, the contact angle of lignocellulose film can be increased by 66.2%, the UV absorption rate can reach 100%, and the oxygen transmission coefficient has no significant effect. In this paper, a new kind of biological packaging material with high oxygen resistance, strong ultraviolet absorption, and water resistance was prepared by recycling waste wheat straw, it has a broad application prospect in the industrial production of packaging materials. Abstract Background MKI67 plays a vital role in the tumour microenvironment (TME) and congenital immunity. The present work focuses on exploring the prognosis prediction performance of MKI67 and its associations with T cell activity and immune infiltration within numerous cancers, especially hepatocellular liver carcinoma (LIHC). Methods Oncomine, GEPIA2, and HPA were adopted to analyse MKI67 levels in different types of cancers. The prognostic prediction performance of MKI67 was evaluated through the TCGA portal, GEPIA2, LOGpc, and Kaplan–Meier Plotter databases. The associations of MKI67 with related gene marker sets and immune infiltration were inspected through TISIDB, GEPIA2, and TIMER. We chose MKI67 to analyse biological processes (BPs) and KEGG pathways related to the coexpressed genes. Furthermore, the gene–miRNA interaction network for MKI67 in liver cancer was also examined based on the miRWalk database. Results MKI67 expression decreased in many cancers related to the dismal prognostic outcome of LIHC. We found that MKI67 significantly affected the prognosis of LIHC in terms of histology and grade. Increased MKI67 levels were directly proportional to the increased immune infiltration degrees of numerous immune cells and functional T cells, such as exhausted T cells. In addition, several critical genes related to exhausted T cells, including TIM-3, TIGIT, PD-1, LAG3, and CXCL13, were strongly related to MKI67. Further analyses showed that MKI67 was associated with adaptive immunity, cell adhesion molecules (CAMs), and chemokine/immune response signal transduction pathways. Conclusion MKI67 acts as a prognostic prediction biomarker in several cancers, particularly LIHC. Upregulation of MKI67 elevates the degree of immune infiltration of many immune cell subtypes, including functional T cells, CD4+ T cells, and CD8+ T cells. Furthermore, MKI67 shows a close correlation with T cell exhaustion, which plays a vital role in promoting T cell exhaustion within LIHC. Detection of the MKI67 level contributes to prognosis prediction and MKI67 modulation within exhausted T cells, thus providing a new method to optimize the efficacy of anti-LIHC immunotherapy.

This study explores the spatial pattern of Historic Chinese Towns and Cities (HCTC) by using a syntactic approach. The HCTC is an important element of the built environment and exhibits a variety of unique spatial characteristics. Although previous research has been focused on qualitative analysis, a quantitative approach to exploring this issue is scarce, leading to insufficient understanding of the spatial characteristics of HCTC. This study presents a quantitative approach to analyzing the spatial pattern of HCTC by utilizing the space syntax method. Four well-preserved historic towns were selected as case studies, each representing a typical spatial type of historic town in China. A series of mathematical measures from space syntax were used to explore the spatial characteristics of HCTC, facilitating expanded interpretation of traditional Chinese ideologies. Results contribute to a more critical understanding of the spatial pattern of HCTC.

Abstract Polyphenols are the most important phytochemicals in our diets and have received great attention due to their broad benefits for human health by suppressing oxidative stress and playing a protective role in preventing different pathologies such as cardiovascular disease, cancer, diabetes, and obesity. The stability of polyphenols depends on their environments of processing and storage, such as pH and temperature. A wide range of technologies has been developed to stabilize polyphenols during processing. This review will provide an overview of the stability of polyphenols in relation to their structure, the factors impacting the stability of polyphenols, the new products deriving from unstable polyphenols, and the effect of a series of technologies for the stabilization of polyphenols, such as chemical modification, nanotechnology, lyophilization, encapsulation, cold plasma treatment, polyphenol–protein interaction, and emulsion as a means of improving stability. Finally, the effects of cooking and storage on the stability of polyphenols were discussed.

Rodenticides are widely used around the world since the 1950s. In Taiwan, an anti-rodent operation initiated 1977 and became a regular action annually implied by the government until 2014. This anti-rodent operation caused many animals of non-target species being exposed by rodenticides and became an environmental issue. The Black-winged Kite (Elanus caeruleus) is a small-sized diurnal raptor widely distributed in the Old World continent. Since 2000, a newly colonized population of this species occurred in Taiwan. Although the Black-winged Kites may suffer from the threats of rodenticides, the population is still growing and soon became the most abundant raptor in farmlands of Taiwan. Whether the Black-winged Kite accumulates higher anticoagulant rodenticide residues than other raptors are still unclear. In this study, liver samples of Black-winged Kites were collected from 2013 to 2016, when the detected residues of anticoagulant rodenticides increased annually. The concentration of residue rodenticide was above 0.2 ppm among 30% of the detected samples, which is the toxicity threshold concentration of other raptors. In the meanwhile, the lesser ricefield rat (Rattus losea), the most common prey of Black-winged Kites, also extended the survival period after fed on rodenticide. The longer survival days after being poisoned can enhance the predation opportunity of raptors, thus affect the accumulated rodenticides in the raptors. This study demonstrates that the Black-winged Kite has higher concentration of anticoagulant rodenticide than most other raptors, which provide the case that the raptor can quickly accumulate rodenticide residues within a short period of time.

This study presents a quantitative approach to exploring the spatio-functional characteristics of historic Chinese towns (HCTs) from a heritage tourism perspective. In recent years, HCTs have evolved from being resident-oriented to being more tourist-oriented, in part due to their heritage significance for attracting tourists. Spatio-functional qualities of a historic town are essential elements of the town’s urban morphology and of great concern for preservation. Previous studies that discussed this issue often used qualitative descriptions, and only limited studies have systematically explored the spatio-functional qualities of HCTs. Thus, there is presently a lack of understanding around this issue, especially based on rigorous quantitative approaches. This study examines the spatio-functional qualities of HCTs using space syntax, a commonly used method in urban studies that enables measurement of spatial characteristics through mathematical means. Four HCTs with heritage significance, Pingyao, Lijiang, Kulangsu, and Wuzhen, were selected as case studies. The study has examined the role of heritage tourism and the results show that tourist-focused functions tend to distribute and aggregate in the urban core of HCTs. By contrast, cultural relics are freely distributed and not expanded over time. Spatio-functional patterns of the four HCTs were theorised, and the paper concludes with suggestions regarding future land-use optimisation for the four HCTs concerning heritage tourism.

Eye size influences visual acuity, sensitivity, and temporal resolution and is a result of vertebrate adaptation to the environment. The habitats of snake species are diverse, ranging from fossorial, terrestrial, arboreal, to aquatic. They also demonstrate a variety of behavioral and physiological characteristics, such as activity time, feeding patterns, and prey detection. In this study, we comparatively investigated how the relative eye size (i.e., eye diameter vs. head width) associated with the ecological (i.e., habitat), behavioral (i.e., diel activity pattern, foraging strategy), and physiological traits (i.e., the presence of pits), respectively, across six snake families from Taiwan. Among the traits we examined, we found that terrestrial and/or diurnal snakes tended to exhibit the larger relative eye size, indicating the evolutionary responses of eye size to changes in habitat types and activity patterns, respectively, while no evidence of how foraging strategies and the presence of pits affected snake eye size was found. Our findings not only shed light on the adaptive significance of the visual system in diversifying the behaviors and the environments exploited in snakes, but also underline the interactive effects of multidimensional evolutionary attributes (e.g., behavior, ecology, physiology and phylogeny) on the evolution of optimal visual performance.

Marine subsidies are vital for terrestrial ecosystems, especially low-productivity islands. However, the impact of losing these subsidies on the terrestrial food web can be difficult to predict. We analyzed 23 years of survey data from Orchid Island to assess the consequences of the abrupt loss of an important marine subsidy. After climatedriven beach erosion and predator exclusion efforts resulted in the abrupt loss of sea turtle eggs from the terrestrial food web, predatory snakes altered their foraging habitats. This increased predation on other reptile species in inland areas, resulting in population declines in most terrestrial reptile species. Comparisons with sea turtle-free locations where lizard populations remained stable supported these findings. Our study emphasizes the cascading effects of generalist predators and the unintended consequences of single-species conservation, highlighting the importance of understanding species interconnectedness and considering potential ripple effects in marine-dependent insular ecosystems.
